Mycorrhiza formation is accompanied by the up- and down regulation of numerous plant genes. The resulting patterns of differential gene expression can be investigated by following accumulation of the respective transcripts or proteins. Targeted approaches use homologous and heterologous DNA probes, PCR fragments for Northern blot analysis or for in situ hybridization, and antibodies for immunocytochemistry. These tools are available because the corresponding genes already have been analyzed for their structure or function. In contrast, non-targeted approaches such as differential RNA display, differential screening and subtractive hybridization, or PAGE and 2D-gel electrophoresis are directed to the identification of new unknown mycorrhiza-regulated genes.
