ABSTRACT: The hypothesis that vesicular-arbuscular mycorrhizae (VAM) could increase phosphorus (P) availability to plants by solubilizing inorganic P or mineralizing organic P was investigated. Solubilization could occur if mycorrhizae or their associated microflora produce siderophores. Siderophores can chelate iron from iron-phosphate complexes, releasing plant available P.
The effect of EDDHA, a synthetic iron chelating compound, on growth of mycorrhizal and nonmycorrhizal big bluestem plants was compared. Mycorrhizal plants in EDDHA-amended soil had significantly greater growth and P uptake than mycorrhizal plants in unamended soil, but EDDHA amendment did not affect growth of nonmycorrhizal plants. If siderophore production occurs in the mycorrhizosphere, then P availability in low pH soils could be significantly increased, and this would
be a feasible mechanism by which mycorrhizal plants could access P sources unavailable to nonmycorrhizal plants.
Mineralization of organic P is another important soil process by which mycorrhizae could increase nutrient availability to plants. The ability of VAM fungus to increase P uptake and plant growth in artificial growth medium or native soil amended with organic P forms was studied. Nonmycorrhizal plant growth and P uptake was improved by two of the individually amended organic P sources and a mixture of all organic P sources, while mycorrhizal plants benefitted from amendment by each of the P sources. Significant uptake of $sp{32}$P from the organic $sp{32}$P incorporated into native prairie soil was evident only in mycorrhizal plants, which were larger and had higher total P uptake than nonmycorrhizal
plants. This suggests that mycorrhizal plants can obtain P from normally unavailable organic P sources. A different approach, isotopic dilution, can be used to assess the role of VAM fungi in mineralization of native soil organic P. However, the approach requires that soluble inorganic $sp{32}$P ($sp{32}$Pi) be
determined without interference from soluble organic $sp{32}$P ($sp{32}$Po). Therefore, a method was developed to partition $sp{32}$Pi and $sp{32}$Po into physically separate solutions before radiation counting from the $sp{32}$Pi. The P separation procedure described in our study provides a methodology for partitioning Pi and Po in soils. This method resulted in complete separation of pure inorganic and organic P sources as well as soil P, and will be suitable for P mineralization experiments.
